烟属野生种主要植物学性状及黑胫病抗性评价

  摘 要 鉴定了烟属植物野生种生育期等植物学性状特性,评价了供试材料对黑胫病(0号及1号生理小种)的抗性,为开展远缘杂交花期相遇及亲本选配提供了基础数据。在玻璃温室内,本研究针对66份烟属植物野生种的生育期等7个植物学性状的调查,检测了烟碱及钾含量,并对黑胫病(0号及1号生理小种)抗性进行了评价。结果表明:供试材料播种到开花时间(DF)为35.00~357.00 d,平均121.67 d;种子千粒重(TSW)为0.02~0.20 g,平均0.09 g;株高(PH)为11.20~292.75 cm,平均119.19 cm;花冠长度(CL)为1.22~11.50 cm,平均3.98 cm;花冠直径(CD)0.67~7.80 cm,平均2.37 cm;最大叶长(MLL)为6.79~61.15 cm,平均25.17 cm;最大叶宽(MLW)为0.65~35.51 cm,平均11.84 cm。烟碱含量为0.03%~1.81%,平均0.34%;钾含量为0.32%~7.11%,平均3.56%。筛选到N. alata PI42334等11份资源同时抗黑胫病0及1号生理小种。

  Abstract The morphological variation of 66 Nicotiana accessions from 50 species were assessed based on 7 morphological traits. The leaf nicotine and potassium concentration of fresh tobacco leaf by kill-enzyme torrefaction method were measured. The evaluation of the accessions for their resistance to Phytophthora parasitica var. nicotianae(race 0 and race1)was conducted. The results showed that the days from sowing to flower(DF)were observed from 35.00 to 357.00 d with an average of 121.67 d. The 1000-seed weight(TSW)varied from 0.02 to 0.20 g with an average of 0.09 g. The plant height(PH)ranged from 11.20 to 292.75 cm with an average of 119.19 cm. The corolla length(CL)ranged from 1.22 to 11.50 cm with an average of 3.98 cm. The corolla diameter(CD)varied from 0.67 to 7.80 cm with an average of 2.37 cm. The maximum leave length(MLL)varied from 6.79 to 61.15 cm with an average of 25.17 cm, while the maximum leave width(MLW)varied from 0.65 to 35.51 cm with an average of 11.84 cm. The leaf nicotine concentration was measured from 0.03% to 1.81% with an average of 0.92%, potassium concentration measured from 0.32% to 7.11% with an average of 3.56%. Eleven wild tobacco accessions were resistant to black shank race 0 and race 1 after black shank disease inoculation. This work provided an important contribution to help breeders in the future on the direction of interspecific crossing attempts.

  烟属种质资源(Genus Nicotiana)包含76个种, 除人工驯化栽培的普通烟草(Nicotiana(本文来自:WwW.BdfqY.Com 千叶帆文摘:烟属野生种主要植物学性状及黑胫病抗性评价) tabacum L.)和黄花烟草(N. rustica L.)外,其余均为烟属野生种质资源[1](简称烟草野生种)。
  黑胫病是土传病害,主要为害烟草根及茎部,是中国烟叶生产的主要病害,每年造成近1亿元的损失[2]。而烟草野生种拥有抗烟草黑胫病、TMV等主要病害、耐旱等逆境胁迫及具有特殊香气等优良性状[3-4],因此充分鉴定、挖掘并利用烟草野生种的优良基因,成为培育优质、抗病品种的关键。
  开展烟草野生种植物学性状分析,是充分利用野生资源前提和基础。20世纪50年代,Goodspeedii等[5]学者开展了烟草野生种分类学研究后,之后一些学者从细胞学及分子生物学方法,开展了烟草起源与进化方面的研究[6]。随着学者对资源调查的深入,陆续有新的烟草野生种被发现[7]。前人多侧重研究烟属植物进化及亲缘关系分析,涉及到的植物学性状也仅为零星几个物种,50份以上的烟草野生种在相同气候环境下的植物学性状研究未见报道。另外,来自蓝茉莉烟草(N. plumbaginifolia)的高抗黑胫病(0号生理小种)Ph基因,已在烟叶生产得到应用和推广[8],但随着抗病品种的推广,出现了黑胫病的优势生理小种转变快等问题[9],严重威胁烟叶生产的可持续发展。
